Approaches to cell culture propagation of hepatitis A virus (HAV) have used either acute infection by passage of infected cell lysates or supernatants into uninfected cells or the passage of persistently infected cells. The findings presented here d monstrate that the growth and recovery of purified virus from foetal rhesus monkey kidney (FRhK4) cells persistently infected with HAV isolate HAS-15 decreased over a 2 to 3 month period. In contrast, high multiplicity acute infection of FRhK4 cells with purified HAS-15 HAV resulted in degeneration f the cell monolayer 2 to 3 weeks later. Large scale propagation of acutely infected cells followed by traditional picornavirus purification procedures reproducibly yielded milligram amounts of purified virus. The effect of interferon (IFN) on infection and maintenance of persistent infection of Borna disease (BD) virus in cell cultures was investigated. Acutely BD virus-infected primary rabbit brain and rat lung cells produced significant levels of interferon detectable 3 days post-infection in the culture supernatants. Rat brain and rat lung cells persistently infected with BD virus produced only moderate levels of IFN over a long period. In contrast, persistently infected Madin-Darby canine kidney (MDCK) cells did not produce detectable amounts of IFN. Exogenous homologous IFN completely inhibited the expression of BD virus antigen in acutely infected rabbit brain cells, when added during the first 24 h after infection. IFN added later (2 to 6 days post-infection) reduced virus titres to different degrees depending on the onset of treatment. However, IFN added to persistently infected rat lung cells did not appear to influence the degree or quality of BD virus antigen expression or the intracellular amount of infectious virus. Two facts indicate that IFN is not involved in the establishment or maintenance of persistent BD virus infection in ritro. Thus, MDCK cells, which could not be induced to produce IFN, can be readily persistently infected with BD virus in vitro, and exogenous

The duration and level of virus excretion in blood and faecesof patientswithhepatitisAvirus (HAV) infection were studied in relation to levels of alanine aminotransferase (ALT), disease severity and HAV genotype. Clinical data, blood and faeces were collected from 27 patients with acute hepatitis A (median age: 33 years) for amaximum of26weeks.Singleblooddonations from55other patients with acute HAV (median age: 32 years) were also used. Virus loads were quantified by competitive nested RT-PCR. HAVwas excreted in faeces for a median period of 81 days after disease onset, with 50 % of patients still excreting high levels at Day 36 (2 106–2108 copies/ml faeces suspension).
