Zwang et al. have identified outputs of two EGF pulses that commit cells to cycle. The first induces components for lipid biosynthesis and sets up an inhibitory latch through p53. The second works through ERK to EGR, and releases the latch to promote Restriction point traverse. Orderly regulation of cell division is an existential requisite for multicellular organisms, and is managed by exposure of cells to growth factor hormones. Despite the wealth of information on transduction of growth factor signals, the interpretation of these signals in the context of commitment to the cell cycle is poorly understood. The report by Zwang and co-workers [$] in the current issue identifies limiting processes induced by pulsed growth factor stimuli, and reveals a role for p53 in “digitization ” of graded inputs to ensure decisive commitment to S. Mammalian cell cultures are maintained in medium supplemented with serum, and will generally arrest in a quiescent state (Go) under conditions of serum deprivation. Once serum is restored, cells proceed to S phase after a characteristic delay (12 to 14 hours in Balb/c 3T3 cells). Nutritional starvation for amino acids or phosphate induces a similar quiescent state,
