Familial adenomatous polyposis coli (FAP) is an autosomal dominant disease characterised by the presence of 100 or more colorectal adenomatous polyps and the inevitable development, within 10 years, of colon cancer if left untreated. Mutations in the adenomatous polyposis gene {APC) whose mode of action is as a tumour suppressor (1), are primarily responsible for the development of the disease and for many sporadic colon cancers. In 1991, the APC gene was located to chromosome 5q21 (2,3) and was subsequently cloned and sequenced (4-7). The transcribed sequence covers 8535 nucleotides and includes 15 exons. The 15th exon is unusually large with 6500 nucleotides, representing 75 % of the total size of the transcript. More than 60 germ line mutations have been identified in the APC gene (8) with significant clustering in the 5 ' half of exon 15 (9). In this study, we have identified four novel mutations in exon
