This article describes the structure of the adolescent romantic and sexual network in a population of over 800 adolescents residing in a midsized town in the midwestern United States. Precise images and measures of network structure are derived from reports of relationships that occurred over a period of 18 months between 1993 and 1995. The study offers a comparison of the structural characteristics of the observed network to simulated networks conditioned on the distribution of ties; the observed structure reveals networks characterized by longer contact chains and fewer cycles than expected. This article identifies the micromechanisms that generate networks with structural features similar to the observed network. Implications for disease transmission dynamics and social policy are explored.

A central idea in marketing and diffusion research is that influentials—a minority of individuals who influence an exceptional number of their peers—are important to the formation of public opinion. Here we examine this idea, which we call the “influentials hypothesis,” using a series of computer simulations of interpersonal influence processes. Under most conditions that we consider, we find that large cascades of influence are driven not by influentials, but by a critical mass of easily influenced individuals. Although our results do not exclude the possibility that influentials can be important, they suggest that the influentials hypothesis requires more careful specification and testing than it has received.

While questions about social cohesion lie at the core of our discipline, no clear definition of cohesion exists. We present a definition of social cohesion based on network connectivity that leads to an operationalization of social embeddedness. We define cohesiveness as the minimum number of actors who, if removed from a group, would disconnect the group. This definition generates hierarchically nested groups, where highly cohesive groups are embedded within less cohesive groups. We discuss the theoretical implications of this definition and demonstrate the empirical applicability of our conception of nestedness by testing the predicted correlates of our cohesion measure within high school friendship and interlocking directorate networks. Anti-viral drug treatment of infections with the human immunodeficiency virus type 1 (HIV-1) usually leads to a rapid decline in the abundance of plasma virus. The effect of single drug therapy, however, is often only short-lived as the virus readily develops drug-resistant mutants. In this paper we provide analytic approximations for the rate of emergence of resistant virus. We study the decline of wildtype virus and the rise of resistant mutant virus in different compartments of the virus population such as free plasma virus, cells infected with actively replicating virus, long-lived infected cells and cells carrying defective provirus. The model results are compared with data on the rise of drug-resistant virus in three HIV-1 infected patients treated with neverapine (NVP). We find that the half-life of latently infected cells is between 10 and 20 days, whereas the half-life of cells with defective provirus is about 80 days. We also provide a crude estimate for the basic reproductive ratio of HIV-1 during NVP therapy. � 1997 Academic Press Limited 1.

The purpose of time series analysis via mechanistic models is to reconcile the known or hypothesized structure of a dynamical system with observations collected over time. We develop a framework for constructing nonlinear mechanistic models and carrying out inference. Our framework permits the consideration of implicit dynamic models, meaning statistical models for stochastic dynamical systems which are specified by a simulation algorithm to generate sample paths. Inference procedures that operate on implicit models are said to have the plug-and-play property. Our work builds on recently developed plug-and-play inference methodology for partially observed Markov models. We introduce a class of implicitly specified Markov chains with stochastic transition rates, and we demonstrate its applicability to open problems in statistical inference for biological systems. As one example, these models are shown to give a fresh perspective on measles transmission dynamics. As a second example, we present a mechanistic analysis of cholera incidence data, involving interaction between two competing strains of the pathogen Vibrio cholerae. 1. Introduction. A

Many recent large-scale studies of interaction networks have focused on networks of accumulated contacts. In this paper we explore social networks of ongoing relationships with an emphasis on dynamical aspects. We find a distribution of response times (times between consecutive contacts of different direction between two actors) that has a power-law shape over a large range. We also argue that the distribution of relationship duration (the time between the first and last contacts between actors) is exponentially decaying. Methods to reanalyze the data to compensate for the finite sampling time are proposed. We find that the degree distribution for networks of ongoing contacts fits better to a power-law than the degree distribution of the network of accumulated contacts do. We see that the clustering and assortative mixing coefficients are of the same order for networks of ongoing and accumulated contacts, and that the structural fluctuations of the former are rather large. Mathematical modeling of viral replication dynamics, based on sequential measurements of levels of virion-associated RNA in plasma during antiretroviral treatment, has led to fundamental new insights into human immunodeficiency virus type 1 pathogenesis. We took advantage of the simian immunodeficiency virus (SIV)-infected macaque model to perform detailed measurements and mathematical modeling during primary infection and during treatment of established infection with the antiretroviral drug (R)-9-(2-phosphonylmethoxypropyl)adenine (PMPA). The calculated clearance half-life for productively infected cells during resolution of the peak viremia of primary infection was on the order of 1 day, with slightly shorter clearance half-lives calculated during PMPA treatment. Viral reproduction rates upon discontinuation of PMPA treatment after 2 weeks were approximately twofold greater than those obtained just prior to initiation of treatment in the same animals, likely reflecting accumulation of susceptible target cells during treatment.
