We describe the basic assumptions and key results of loop quantum gravity, which is a background independent approach to quantum gravity. The emphasis is on the basic physical principles and how one deduces predictions from them, at a level suitable for physicists in other areas such as string theory, cosmology, particle physics, astrophysics and condensed matter physics. No details are given, but references are provided to guide the interested reader to the literature. The present state of knowledge is summarized in a list of 35 key results on topics including the hamiltonian and path integral quantizations, coupling to matter, extensions to supergravity and higher dimensional theories, as well as applications to black holes, cosmology and Plank scale phenomenology. We describe the near term prospects for observational tests of quantum theories of gravity and the expectations that loop quantum gravity may provide predictions for their outcomes. Finally, we provide answers to frequently asked questions and a list of key open problems.

Factors that affect he probability of genetic transformation f Escherichia coli by plasmids have been evaluated. A set of conditions is described under which about one in every 400 plasmid molecules produces a transformed cell. These conditions include cell growth in medium containing elevated levels of Mg 2+. and incubation of the cells at 0 ~ in a solution of Mn 2+, ("a 2+, Rb + or K +, dimethyl sulfoxide, dithiothreitol, and hexamine cobalt (III). Transibrmation efficiency declines linearly with increasing plasmid size. Relaxed and supercoiled plasmids transfol'm with similar probabilities. Nontransforming DNAs compete consistent with mass. No significant variation is observed between competing DNAs of difi~rent source, complexity, length or form. Competition with both transforming and nontransforming plasmids indicates that each cell is capable of taking up many DNA molecules, and that the establishment of a transformation event is neither helped nor hindered significantly by the presence of multiple plasmids. 1. We describe a hardware and software system for digitizing the shape and color of large fragile objects under nonlaboratory conditions. Our system employs laser triangulation rangefinders, laser timeofflight rangefinders, digital still cameras, and a suite of software for acquiring, aligning, merging, and viewing scanned data. As a demonstration of this system, we digitized 10 statues by Michelangelo, including the wellknown figure of David, two building interiors, and all 1,163 extant fragments of the Forma Urbis Romae, a giant marble map of ancient Rome. Our largest single dataset is of the David  2 billion polygons and 7,000 color images. In this paper, we discuss the challenges we faced in building this system, the solutions we employed, and the lessons we learned. We focus in particular on the unusual design of our laser triangulation scanner and on the algorithms and software we developed for handling very large scanned models.
